Scheme of RNA transcription in calicivirus-infected cells.

چکیده
Feline calicivirus (FCV) is a member of the Caliciviridae and contains a positive-stranded, non-segmented. polyadenylated RNA genome. It codes for at least one structural protein [ 1. 21 and one nonstructural protein [3]. The mode of synthesis of the viral RNA is still not clear. Neil and Mengeling identified three subgenomic RNAs in addition to the full length genome [4]. However, Carter reported that the number of virus specific RNAs detectable in infected cells was 8, and showed that RNA species 1 to 5 were represented as negative strands which may serve as templates in the synthesis of these RNAs [5]. Neil and Mengeling also reported the existence of a 3' co-terminal nested set of subgenomic RNAs in common with other virus families [4].

RNA labeled with [3H]uridine from Vero cells infected with San Miguel sea lion virus in the presence of actinomycin D was analyzed by glycerol density gradient sedimentation and polyacrylamide gel electrophoresis. The predominant single-stranded RNA (36S, 2.6 x 10(6) molecular weight) was genome size. There was also a prominent 22S, 1.1 x 10(6)-molecular weight, single-stranded component and on...

Mycobacterium aviumsubspecies paratuberculosis (MAP), as an obligate intracellular bacterium, causes paratuberculosis (Johne’s disease) in ruminants. Plus, MAP has consistently been isolated from Crohn’s disease (CD) lesions in humans; a notion implying possible direct causative ...

A membranous fraction that could synthesize viral RNA in vitro in the presence of magnesium salt, ribonucleotides, and an ATP-regenerating system was isolated from feline calicivirus (FCV)-infected cells. The enzymatically active component of this fraction was designated FCV replication complexes (RCs), by analogy to other positive-strand RNA viruses. The newly synthesized RNA was characterized...
متن کاملDetection of the ORF3 polypeptide of feline calicivirus in infected cells and evidence for its expression from a single, functionally bicistronic, subgenomic mRNA.
Feline calicivirus (FCV) is a small positive-stranded RNA virus within the family Caliciviridae. Its genome is 7690 nucleotides in length and encodes three open reading frames (ORFs). The smallest, ORF3, is located at the extreme 3' end of the genome and can potentially encode a polypeptide of approximately 12 kDa. In this paper, we report the identification of an ORF3-encoded polypeptide in FC...
متن کاملIdentification of a protein linked to the genomic and subgenomic mRNAs of feline calicivirus and its role in translation.
125I protein labelling of oligo(dT)-selected RNA from feline calicivirus (FCV)-infected cells revealed that the genomic and 2.4 kb subgenomic RNAs of FCV are linked to a 15 kDa protein (VPg). Proteinase K treatment of FCV RNA, to remove VPg, led to a decrease in the translatability of the RNA, but there was no obvious change in the site of RNA initiation. Addition of the cap analogue 7-methylGT...
